ISOLATION AND CHARACTERIZATION OF TRITERPENOID COMPOUNDS
FROM STEM BARK OF Chisocheton macrophyllus

Background: C. macrophyllus is one of the species of the Chisocheton genus. The triterpenoid
compounds contained in the stem bark of C. macrophyllus from Sukabumi have not been
reported yet. Previous studies have shown that triterpenoid compounds, damara-20,24-dien-3-
on, have been isolated from the stem bark of C. macrophyllus from Bogor Botanical
Gardens. Therefore, this study aims to isolate and identify triterpenoid compounds contained
in the stem bark of C. macrophyllus from Sukabumi.

Methodology: The extraction of C. macrophyllus was carried out using maceration with methanol. The extract was fractionated using a separating funnel and produced ethyl acetate and n- hexane fraction. The ethyl acetate fraction was separated and purified using vacuum liquid chromatography and column chromatography. The isolate was identified using 1H-NMR, 13C-NMR, HSQC, and HMBC spectroscopy.

Results: The isolate isolated from the stem bark of C. macrophyllus has a white powder and its amount was 6 mg. 13C-NMR spectrum showed that the isolate had 30 carbon signals and 1H-NMR spectra showed 22 proton signals. Based on the interpretation of the 1H-NMR, 13C-NMR, HSQC, HMBC, and literature spectrum, isolate has 4 cyclic skeletons that are predicted to be dammarane or protostanes.

Conclusion: The triterpenoid compound was isolated from the stem bark of C. macrophyllus
from Sukabumi was dammarane or protostane triterpenoid.
