Abstract

Introduction : the TLC profile with mobile phase variations can be used as a preliminary study for the selection of column chromatography methods for the isolation of compounds in extracts.Method: The lemon peel was macerated with methanol, then fractionated with chloroform. Methanol and chloroform fractions were analyzed by TLC stationary phase silica gel GF 254 mobile phase combination n-hexane : ethyl acetate 9:1 to 1:9 with UV 254, citroborate and AlCl3 as spot viewer. Results : Chalcone began to elude at n-hexane: ethyl acetate 6:4 with Rf 0.320 increasing with the addition of the percentage of ethyl acetate. The first chalcone has passed the elution limit at n-hexane: ethyl acetate 6:4 while the second still exists up to n-hexane: ethyl acetate 1:9 with Rf value 0.057; 0.257; 0.485 and 0.857.

Conclusions : The TLC profile can be used as a reference for the isolation of chalcone with KVC using the mobile phase combination of n-hexane: ethyl acetate gradient from 6:4 to 1:9.
